Moderna and Merck announce breakthrough, could usher in wave of cancer vaccines

Moderna and Merck have announced a significant breakthrough in cancer treatment, with their personalized mRNA cancer vaccine, intismeran (also known as V940 or mRNA-4157), demonstrating success in a late-stage clinical trial. The Phase 3 INTerpath-001 study, which combined intismeran with Merck's immunotherapy Keytruda, met its primary endpoints by significantly extending recurrence-free survival and reducing the spread of cancer in patients with high-risk melanoma following surgical removal of their tumors. This marks a pivotal moment, as it is the first neoantigen cancer vaccine to succeed in such a rigorous late-stage trial.

The individualized vaccine operates by analyzing each patient's tumor to identify unique genetic mutations, or neoantigens, and then custom-designs an mRNA treatment to train the immune system to specifically target and attack these cancerous cells. While detailed data from the Phase 3 trial are pending release at an upcoming medical meeting, earlier Phase 2 results had indicated a substantial reduction in the risk of melanoma recurrence or death when the combination therapy was used.

This advancement is seen by many as ushering in a new era for cancer care, particularly in the realm of personalized medicine. Both companies are preparing to present the full trial data and will engage with regulatory bodies to pursue potential accelerated approval for the melanoma treatment. Furthermore, the promising results have fueled optimism for expanding this mRNA vaccine platform to address other cancer types, including lung, kidney, and bladder cancers.
